I think there are many factors at play, but my guess is these three play a big role:

1 - We have polluted our world to such a degree that our life-sustaiining environment is now a chemical cocktail/soup (air, water, land, and the animals inhabiting it). Industry has contaiminated environmental systems to a point where populations can no longer live and/or reproduce (plenty of sources.. ie intersex amphibians and fish due to pharmaceuticals in the water).
2 - Climate change. Conditions are changing too quickly for species to adapt. (ie species of frogs that usually hibernate by freezing over the winter are dying because the temperatures aren't cold enough)
3 - Loss of habitat = ripple effect through ecosystems. Mass extinctions as food-chains collapse.
