Youth serum: Young blood rejuvenates old cells, and now researchers are working to determine which factors within are responsible for the regenerative boost
Young blood rejuvenates old muscles in mice, making compounds within a target for regenerating various tissues affected by aging.

Old muscles can repair themselves but for some reason do not. In young mice and humans, muscle stem cells called satellite cells come to the rescue of damaged muscle. In old muscle the cells are present but they do not respond in this way.

Thomas Rando at Stanford University School of Medicine in California and colleagues have now shown that the blood bathing the cells, rather than the cells themselves, may be at fault.

The researchers attached old mice to younger mice in a way that caused them to share a blood supply. They then induced damage in the muscles of the older mice.

In the presence of young blood, the old muscles healed normally. The same effect wasn´t seen when the mice were attached to other old mice.

Furthermore, looking at the satellite cells, Rando and colleagues found that old muscles bathed in young blood ramped up production of a protein called Delta in response to injury, while normally satellite cells in old blood maintain the same pre-injury levels of the protein even after muscle damage.

When the satellite cells from old and young mice were studied in a lab dish, blood serum from old mice caused the cells from young mice to decrease Delta production and divide less frequently while blood serum from young mice caused the cells from old mice to increase Delta production and divide.

Important niche

"We need to consider the possibility that the niche in which stem cells sit is as important in terms of stem cell aging as the cells themselves," says Rando.

In other words, the mix of chemicals surrounding the cells rather than the cells themselves may explain reduced regenerative capacity.

Support for this came from similar work in which Rando and colleagues found that the liver of old mice connected to younger mice regenerated more robustly than would normally be expected.

Rando says that age-related inability to repair many tissues, from liver to brain, may depend on the environment of regenerative cells rather than just the cells themselves.

The results don´t mean that blood transfusions will become a viable option for fighting aging. For one thing, the mice were genetically altered to prevent immune complications. For another, they shared a blood supply for six weeks.

The goal now is to find what factors in blood are responsible for the rejuvenative effect—no small task because blood contains thousands of proteins, lipids, sugars and other small molecules. Additionally, different chemicals may restore regenerative capacity to different types of cells.

"Another approach is to pick factors that are good candidates and see if any of them or some combination recapitulate the effect of the younger blood," says Rando.
