The ability to assemble nanoparticles into arrays, networks, and circuits in a precise and controlled manner is key to the fabrication of a variety of nanodevices. Networks of nanometer-sized metal or semiconductor islands, or quantum dots, may exhibit a variety of quantum phenomena, with applications in optical devices, nanometer-sized sensors, advanced computer architectures, ultra dense memories, and quantum-information science and technology. The challenge is that fabrication with nanoscale precision of nanoparticle arrays in a time and cost effective manner remains a formidable task. Interest in the concept of self-assembled nanostructures led to the idea of using DNA as a scaffold or template for the programmed assembly of nanoscale arrays. DNA can be modified with functional groups at predetermined sites to allow for the attachment of other molecules in a specific manner. We have designed and demonstrated a new approach for binding nanoparticles to DNA. Functionalized nanoparticles are covalently bound to internal, chemically modified bases on double-stranded DNA without the presence of destabilizing "nicks" along the DNA backbone. In addition, we report an approach for thiolating one end of the DNA/nanoparticle product and attaching it to a gold surface. The ability to attach one or both ends of the DNA/gold complex, after generation of the desired pattern, to fixed contacts or electrodes is necessary for nanodevices fabrication.

DNA oligonucleotides were designed with amino-modified bases for attachment to carboxylic acid functionalized gold particles. Two double-stranded DNA sequences were used for binding nanoparticles. Sequence 1 DNA was 22 base pairs long with two binding sites for gold per DNA molecule. The separation between gold binding sites was 3.7 nm. Sequence 2 DNA was 30 base pairs long, had one gold binding site per DNA molecule, and, after ligation, a 10.5 nm separation between binding sites. For AFM imaging, the DNA was ligated to produce longer molecules that would be easier to image. Gold nanoparticles with two different passivating coatings were tested. Particles with an average diameter of 1.5 nm were synthesized with a mercaptosuccinic acid coating, and particles approximately 2.5 nm in size were coated with thioctic acid Each particle has multiple reactive carboxyl groups on its surface. In order to decrease the probability for one particle binding to many amino groups on the DNA, methylamine was used to block some of the carboxyl groups on the gold. Methylamine was chosen for this purpose because of its small size and similarity to the methylene side chain containing the amino group on the DNA. The reaction between the amino group on the DNA and the carboxyl group on the gold particle was facilitated using a standard chemical method for joining carboxyl groups to amino groups. Analysis of the products by gel electrophoresis and atomic force microscopy (AFM) showed the gold particles bound to the DNA. In addition, absorbance spectra of the gold nanoparticles in the presence of DNA provide evidence of binding. This technique addresses a basic need to assemble nanometer-scale objects in a programmable manner and in a parallel fashion, from the bottom up.

This is so weird that this was posted (I must have missed it earlier) because I was JUST reading about spontaneous DNA on the Russian Space Station. Another scientist supposedly discovered complex DNA organisms by putting intense heat on silica sand in a vacuum. What is sand? CRYSTAL!!!

In reading up on the similarities between space and the deep sea (very VERY similar), I wonder if these sailors didn't experience some sort of depressurization. I would assume they would know the dangers of that possibility if they are on submarines, but it seems very peculiar.

Thread: Desperate actions in Gulf of Mexico Oil Spill disaster, Experimental Oil eating bacteria/microbes, causing unexpected results.

At this point it's unclear how much of an environmental threat oil spreading from the BP spill will cause, but the federal government is mobilizing thousands of workers to prepare for the worst. They have a potential ally: microbes that have evolved an ability to break down oil that seeps from the ocean bottom. It gets devoured by a variety of bacteria, which eat it by chemically transforming its compounds into useful cellular constituents. "If it wasn't for the natural ability of bacteria to eat oil we would all be knee-deep in the stuff," says bioremediation expert Ken Lee of the Bedford Institute of Oceanography in Nova Scotia, Canada.

So could bugs help cleanse the gulf? A number of companies have tried to create bacteria that could break down oil on demand, but Lee and colleague Albert Venosa of the Environmental Protection Agency say that experiments have shown that novel bacteria, even if they show promise in the lab, cannot compete with bacteria already living on beaches and marshes. Experiments have shown that adding nutrients like nitrogen, phosphorus, and potassium to the beaches can speed up the ability of natural bacteria to break down oil. "What would've taken 5 or 6 years to accomplish can occur in a single summer," says Lee.

While adding such fertilizers has worked in small scale coastal experiments in which oil was purposefully spread on wetlands, experts don't know of examples from an actual spill. The challenge with wetland marshes is that the toxicity of the oil can kill plants before the microbes have a chance to get to work on the oil. "If that happens, you can lose the whole marsh," Lee says. Workers have been trying to remove as much oil at sea as possible to reduce the amount that hits the shore. They will also need to deploy protective booms to protect the wetlands.

Another challenge: it appears to Vennosa, from photos and news reports, that the oil leaking from the borehole contains water. That makes it more difficult to burn at sea, more difficult to break up into tiny particles using chemicals called dispersants, and more difficult for bacteria to break down. But he cautions that he has not gotten real data on the makeup of the oil striking the coast.

I'm following this thread from the beginning.

One thing keeps bothering me:

150 vessels at spot, so lets say at least 1000 or more personel.

Ships are going from the site to harbour -->contamination?

those ROV's coming to surface-->contamination, yes or no

vessels--> engines bacteria eats oil---> engine will stop

In short: how come that this little village of vessels is still running like a well organised unit ?

Can anyone comment on this, because I cannot put these in the whole picture.

Great thread, learned a lot about new technologies.

In church tonight, prayed for the people of the GOM, for those who are working on that damn thing this night, for...everyone and everything in that region.

Sometimes fear can be the worst thing in the world because it disrupts our thoughts and the lack of peace is almost painful. But don't let this fear rob you of that. ......I do have a comment about the Bermuda Triangle however.I have read that when Atlantis sank she took an ENORMOUS CRYSTAL with her.On certain days when the angle of the sun is just right the crystal heats the methane hydrate and releases HUGE amounts of METHANE cAusing the immediate sinking of any ship or aircraft unlucky enough to be in the area!If Atlantis is rising you can bet this crystal is rising with it!I would imagine that the USA would not want anyone even taking photos of it!We may be talking about a photo here not a rogue virus!Let's see what Krispy and everyone else comes up with tonight!Sweet Dreams xx

I'm following this thread from the beginning.

One thing keeps bothering me:

150 vessels at spot, so lets say at least 1000 or more personel.

Ships are going from the site to harbour -->contamination?

those ROV's coming to surface-->contamination, yes or no

vessels--> engines bacteria eats oil---> engine will stop

In short: how come that this little village of vessels is still running like a well organised unit ?

Can anyone comment on this, because I cannot put these in the whole picture.

Great thread, learned a lot about new technologies.

and please, you all, stay at the positive side.
