A private investigator will find out real quick if a spouse is cheating. They are commonly hired for this very reason.

I would tell your brother what you saw and speak with him about his options to further investigate the matter. Maybe he'll want to speak with his wife about it, or maybe he'll want to investigate a little further.

I think OP cares about his family and his intuition is telling him that something is 'amiss' with regards to his brother's marriage. His brother already made a comment about it, his brother is away for long stretches of time, and he sees his sister-in-law embracing another unknown man with a kiss on the cheek while her husband is away. Anyone who doesn't think this situation deserves more investigation is operating with their eyes wide shut.
